Yeah it's kind of a weird situation. I have done some testing and noticed that if you only use the Transform Constraint for rotation it works fine, but the translation doesn't work for some reason.
A workaround is to use the constraint for easier animation in Spine and then keyframe the translation of the affected bones afterwards.
